  • Amlodipine is a dihydropyridine calcium antagonist (calcium ion antagonist or slow-channel blocker) that inhibits the transmembrane influx of calcium ions into vascular smooth muscle and cardiac muscle. Amlodipine is a peripheral arterial vasodilator that acts directly on vascular smooth muscle to cause a reduction in peripheral vascular resistance and reduction in blood pressure.
  • Recommended starting dose: 5 mg once daily with maximum dose10 mg once daily. Small, fragile, or elderly patients, or patients with hepaticinsufficiency may be started on 2.5 mg once daily.
  • Amlodipine tablets are contraindicated in patients with known hypersensitivity to amlodipine.

  • Pregnancy category C,use only if the potential benefit justifies the potential risk to the fetus.Discontinue lactation when administering Amlodipine. Effect on patients less than 6 years old is not known. In geriatric patients start dosing at the low end of the dose range, due to the greater frequency of decreased hepatic, renal, or cardiac function and of concomitant disease or other drug therapy.
  • When Amlodipine and sildenafil were used in combination, each agent independently exerts its own blood pressure lowering effect.Monitor for symptoms of hypotension and edema when amlodipine is co-administered with CYP3A4 inhibitors(e.g., ketoconazole, itraconazole, ritonavir).
  • Most common adverse reactions are headache, pedal edema which occurred in a dose related manner.
  • Anti-Hypertensives
