Fungina Capsules (Fluconazole)

  • Each capsule contains 150mg of fluconazole.
  • For the treatment of systemic candidiasis, mucosal candidiasis, cryptococosis, prevention of fungal infections in patients with malignancy.
  • Fluconazole is a highly selective inhibitor of fungal cytochrome P450 dependent enzyme lanosterol 14-α-demethylase. This enzyme functions to convert lanosterol to ergosterol. The subsequent loss of normal sterols correlates with the accumulation of 14-α-methyl sterols in fungi and may be responsible for the fungistatic activity of fluconazole
  • For vaginal candidiasis 150 mg is adminisstered as a single oral dose.
  • Fluconazole is contraindicated in patients who have shown hypersensitivity to fluconazole or to any of its excipients.Coadministration of other medicinal products known to prolong the QT interval and which are metabolised via the cytochrome P450 (CYP) 3A4 such as cisapride, astemizole, pimozide, quinidine and erythromycin are contraindicated in patients receiving fluconazole.
  • Hepatic injury: Fluconazole should be administered with caution to patients with liver dysfunction. Fluconazole carries the potential of causing anaphylaxis and exfoliative skin disorders. Some azoles, including fluconazole, have been associated with prolongation of the QT interval on the electrocardiogram.Fluconazole should be administered with caution to patients with renal dysfunction.
  • Fluconazole in standard doses and short-term treatments should not be used in pregnancy unless clearly necessary. Breast-feeding is not recommended. Fluconazole is not recommended under 6 months of age.
  • Concomitant administration of fluconazole with amiodarone may increase QT prolongation. Fluconazole has the potential to increase the systemic exposure of the calcium channel antagonists as CCBs are metabolized by CYP3A4 enzymes. Fluconazole increases the systemic exposure of NSAIDs, celecoxib and tofacitinib when administered concomitantly. Prothrombin time may be increased in patients receiving concomitant fluconazole and coumarin-type anticoagulants. Clinically significant hypoglycemia may be precipitated by the use of fluconazole with oral hypoglycemic agents. Fluconazole increases the plasma concentrations of phenytoin and vinca alkaloids.Fluconazole increases the serum concentrations of theophylline.
