Petril Plus LS Tablets (Clonazepam+ Escitalopram )

• Composition :
  Each Petril Plus tablet contains Clonazepam 0.5 mg and Escitalopram 10 mg Petril Plus LS contains Clonazepam 0.25 mg and Escitalopram 10 mg
• Indications :
  For the treatment of comorbid depression and anxiety disorder.
• Pharmacodynamics :
  Clonazepam enhances the activity of gamma aminobutyric acid (GABA), the major inhibitory neurotransmitter in the central nervous system. The mechanism of antidepressant action of escitalopram, the S-enantiomer of racemic citalopram, is linked to potentiation of serotonergic activity in the central nervous system (CNS) resulting from its inhibition of CNS neuronal reuptake of serotonin (5-HT).
• Dosage and administration :
  one tablet once daily, may be taken with or without food
• Contraindications :
  Contraindicated in patients with hypersensitivity to Clonazepam and Escitalopram. In patients with clinical or biochemical evidence of significant liver disease. Concomitant use of monoamine oxidase inhibitors.
• Precautions and warnings :
  Clonazepam-Interference with Cognitive and Motor Performance, Increase risk of suicidal thoughts or behavior. Escitalopram- Increase risk of suicidal thoughts or behavior and serotonin syndrome.
• Use in pregnancy, lactation & pediatrics :
  Pregnancy Category D: Clonazepam has harmful pharmacological effects on pregnancy and foetus/newborn child .Clonazepam is excreted in human milk mothers should not breastfeed their infants, Safety and effectiveness in pediatric patients with panic disorder below the age of 18 have not been established.
• Drug interactions :
  Phenytoin, carbamazepine or phenobarbital alter the pharmacokinetics of Clonazepam. Escitalopram- Concomitant use with SSRIs, SNRIs or Tryptophan is not recommended. Caution should be used when tablets is taken in combination with other centrally acting drugs.
• Adverse reactions :
  nausea, memory impairment, uncoordinated body movements, fatigue and somnolence
