Maxpride Tablets (Amisulpiride).

  • Each tablet contains Amisulpride 50 mg/100 mg/200 mg.
  • Amisulpride is indicated for the treatment schizophrenia.
  • Amisulpride binds selectively with a high affinity to human dopaminergic D2/D3 receptor subtypes whereas it is devoid of affinity for D1, D4 and D5 receptor subtypes. This pharmacological profile explains the clinical efficacy of amisulpride against both negative and positive symptoms of schizophrenia.
  • For acute psychotic episodes, oral doses between 400 and 800 mg are recommended. If required, the daily dose may be increased up to 1200 mg. For patients characterized by predominant negative symptoms, oral doses between 50 mg/d and 300 mg/d are recommended. Doses should be adjusted individually. Amisulpride can be administered once daily at oral doses up to 300 mg, higher doses should be administered bid.
  • Contraindicated in patients with known hypersensitivity to Amisulpride, Concomitant prolactin-dependent tumours, Phaeochromocytoma, use in combination with levodopa.
  • Neuroleptic Malignant Syndrome : elevated CPK may occur amisulpride should be discontinued, appropriate glycemic monitoring in diabetes mellitus, In renal insufficiency the dose should be decreased or intermittent treatment should be considered, Patients with epileptic history should be closely monitored.
  • The use of amisulpride is not recommended during pregnancy and in women of child bearing potential not using effective contraception, unless the benefits justify the potential risks. decision must be made whether to discontinue breast-feeding or to abstain from amisulpride therapy taking into account the benefit of breastfeeding for the child and the benefit of therapy for the woman, Safety and effectiveness in pediatric patients have not been established.
  • Caution should be exercised on concomitant administration with CNS depressant, Antihypertensive drugs and other hypotensive medications and Clozapine, Caution is advised when prescribing amisulpride with medicines known to prolong the QT interval. Amisulpride may enhance the central effects of alcohol.
  • Amisulpride causes an increase in plasma prolactin levels which is reversible after drug discontinuation. This may result in galactorrhoea, amenorrhoea, gynaecomastia, breast pain, and erectile dysfunction. Insomnia, anxiety, agitation.
